The Fras1/Frem gene family encodes for structurally similar, developmentally regulated extracellular matrix proteins. Mutations in Fras1, Frem1 and Frem2 have been identified in different classes of mouse bleb mutants, while defects in the human orthologs FRAS1 and FREM2 are causative for Fraser syndrome. The hallmark phenotypic feature of bleb mice is embryonic skin blistering due to dermal-epidermal detachment. The similarity of the phenotypic characteristics among the bleb mouse mutants, together with the fact that Fras1/Frem proteins are co-localized in embryonic epithelial basement membranes, suggest that they operate in a common pathway. Here, we report for the first time the immunofluorescence pattern of Frem3 and provide a comparative analysis of the spatiotemporal localization of all Fras1/Frem proteins during mouse embryonic development. We demonstrate their overall co-localization in embryonic epithelial basement membranes, with emphasis on areas of phenotypic interest such as eyelids, limbs, kidneys, lungs and organs of the gastrointestinal tract and the central nervous system. We further studied collagen VII, impairment of which produces dystrophic epidermolysis bullosa, a postnatal skin blistering disorder. We show that basement membrane levels of collagen VII rise at late embryonic life, concomitant with descending Fras1/Frem immunolabeling.  相似文献

The R7 family of regulators of G protein signaling (RGS) is involved in many functions of the nervous system. This family includes RGS6, RGS7, RGS9, and RGS11 gene products and is defined by the presence of the characteristic first found in Disheveled, Egl-10, Pleckstrin (DEP), DEP helical extension (DHEX), Gγ-like, and RGS domains. Herein, we examined the subcellular localization of RGS7, the most broadly expressed R7 member. Our immunofluorescence studies of retinal and dorsal root ganglion neurons showed that RGS7 concentrated at the plasma membrane of cell bodies, in structures resembling lamellipodia or filopodia along the processes, and at the dendritic tips. At the plasma membrane of dorsal root ganglia neurons, RGS7 co-localized with its known binding partners R7 RGS binding protein (R7BP), Gαo, and Gαq. More than 50% of total RGS7-specific immunofluorescence was present in the cytoplasm, primarily within numerous small puncta that did not co-localize with R7BP. No specific RGS7 or R7BP immunoreactivity was detected in the nuclei. In transfected cell lines, ectopic RGS7 had both diffuse cytosolic and punctate localization patterns. RGS7 also localized in centrosomes. Structure-function analysis showed that the punctate localization was mediated by the DEP/DHEX domains, and centrosomal localization was dependent on the DHEX domain.  相似文献

A constitutive law that includes three analytical expressions was recently proposed to approximate the low, physiologic, and high-stress parts of the aortic stress-strain relation in uniaxial tension, consistent with the biphasic nature of the aortic wall under passive conditions. This consistency, and the fact that previous phenomenological uniaxial laws have only indirectly been related to vessel wall structure, motivates the investigation of the structural basis underlying the newly proposed three-part constitutive law. For this purpose, longitudinally oriented aortic strips were fixed in Karnovsky's solution, while subjected to various pre-selected levels of uniaxial tensile stress. Light microscopy examination disclosed that the elastic lamellae gradually unfolded at low and were almost straight at physiologic and high stresses, while collagen fibers reoriented in the longitudinal axis at low, started uncoiling at physiologic, and straightened massively at high stresses. In the circumferential sections, the elastic lamellae and the circumferentially distributed collagen bundles remained wavy at all levels of longitudinally applied stress. These microstructural changes suggest that elastin becomes load-bearing at low, and collagen at physiologic but mostly at high stresses, so that the first and third parts of the constitutive law are in turn due to the presence of elastin and collagen alone, and the second due to both elastin and collagen. The structural basis of this constitutive law allows physically significant interpretation of its parameters, offering insight into how the aortic microstructure determines the macromechanical response.  相似文献

Preeclampsia is a pregnancy-related disorder that causes maternal and fetal morbidity and mortality. Its exact inheritance pattern is still unknown, and genome searches for identifying susceptibility loci for preeclampsia have thus far produced inconclusive or inconsistent results. We performed a heterogeneity-based genome search meta-analysis (HEGESMA) that synthesized the available genome scan data on preeclampsia. HEGESMA identifies genetic regions (bins) that rank highly on average in terms of linkage statistics across genome scans (searches). The significance of each bin’s average rank and heterogeneity across scans was calculated using Monte Carlo tests. The meta-analysis involved four genome-scans on general preeclampsia and five scans on severe preeclampsia. In general preeclampsia, 13 bins had significantly high average rank (P rank < 0.05) by either unweighted or weighted analyses, while four of them (2p11.2–2q21.1, 9q21.32–9q31.2, 2p15–2p11.2, 2q32.1–2q35) were formally significant by both analyses. Heterogeneity of bin 2.8 (2q32.1–2q35) was significantly low in both unweighted and weighted analysis (P Q < 0.01). In severe preeclampsia, 10 bins had significantly high average rank by either unweighted or weighted analyses and five of them (3q11.1–3q21.2, 2q37.1–2q37.3, 18p11.32–18p11.22, 2p15–2p11.2, 7q34–7q36.3) were significant by both analyses. Bin 2q37.1–2q37.3 showed marginal low heterogeneity in unweighted and weighted analysis (P Q = 0.06). Results should be interpreted with caution as the p values were modest. Further investigation of these regions by genotyping with additional markers and families may help to direct the identification of candidate genes for preeclampsia.  相似文献

As genome-scale metabolic reconstructions emerge, tools to manage their size and complexity will be increasingly important. Flux balance analysis (FBA) is a constraint-based approach widely used to study the metabolic capabilities of cellular or subcellular systems. FBA problems are highly underdetermined and many different phenotypes can satisfy any set of constraints through which the metabolic system is represented.Two of the main concerns in FBA are exploring the space of solutions for a given metabolic network and finding a specific phenotype which is representative for a given task such as maximal growth rate. Here, we introduce a recursive algorithm suitable for overcoming both of these concerns. The method proposed is able to find the alternate optimal patterns of active reactions of an FBA problem and identify the minimal subnetwork able to perform a specific task as optimally as the whole.Our method represents an alternative to and an extension of other approaches conceived for exploring the space of solutions of an FBA problem. It may also be particularly helpful in defining a scaffold of reactions upon which to build up a dynamic model, when the important pathways of the system have not yet been well-defined.  相似文献

Background

The pandemic by the novel H1N1 virus has created the need to study any probable effects of that infection in the immune system of the host.

Methodology/Principal Findings

Blood was sampled within the first two days of the presentation of signs of infection from 10 healthy volunteers; from 18 cases of flu-like syndrome; and from 31 cases of infection by H1N1 confirmed by reverse RT-PCR. Absolute counts of subtypes of monocytes and of lymphocytes were determined after staining with monoclonal antibodies and analysis by flow cytometry. Peripheral blood mononuclear cells (PBMCs) were isolated from patients and stimulated with various bacterial stimuli. Concentrations of tumour necrosis factor-alpha, interleukin (IL)-1beta, IL-6, IL-18, interferon (FN)-alpha and of IFN-gamma were estimated in supernatants by an enzyme immunoassay. Infection by H1N1 was accompanied by an increase of monocytes. PBMCs of patients evoked strong cytokine production after stimulation with most of bacterial stimuli. Defective cytokine responses were shown in response to stimulation with phytohemagglutin and with heat-killed Streptococcus pneumoniae. Adaptive immune responses of H1N1-infected patients were characterized by decreases of CD4-lymphocytes and of B-lymphocytes and by increase of T-regulatory lymphocytes (Tregs).

Conclusions/Significance

Infection by the H1N1 virus is accompanied by a characteristic impairment of the innate immune responses characterized by defective cytokine responses to S.pneumoniae. Alterations of the adaptive immune responses are predominated by increase of Tregs. These findings signify a predisposition for pneumococcal infections after infection by H1N1 influenza.  相似文献

Submerged hydrophyte vegetation consists of a highly important biotic component of maintaining lake ecosystems towards a “clear water” ecological status. Aquatic macrophytes are well known to play a significant multidimensional role in lakes by competing with phytoplankton growth, stabilising sediment and offering refuge to fishes, macro-invertebrates and littoral zooplankton, amongst others. Zooplanktons that are associated with macrophyte beds, in particular, may act as a positive feedback mechanism that contributes to maintaining a clear-water state. Although there are several studies investigating the relationships between macrophytes and zooplankton in European lakes, few have yet been carried out in Greek lakes. Seasonal field sampling was conducted from spring 2006 to autumn 2008 in four lakes of northwestern Greece. Zooplankton samples were collected from within hydrophyte beds in each lake to estimate their relative abundance and species density. Hydrophyte abundance and composition was recorded on a five-point scale. Moreover, water samples were analysed to determine nutrient and chlorophyll-a concentration. Pearson correlations between zooplankton density and key physicochemical variables were conducted to distinguish significant abiotic variables related with major zooplankton groups. Kruskal–Wallis non-parametric analysis was used to test for significant differences in zooplankton composition and environmental variables amongst the five hydrophyte abundance classes. In addition, Canonical correspondence analysis was used to distinguish possible correlations amongst the macrophyte and zooplankton species. Zooplankton density was significantly higher in dense macrophyte vegetation. Small-sized species (e.g. Rotifera) dominated the zooplankton community, indicating the eutrophic nature of the lakes. Large Cladocera were present in low abundance and were mostly littoral. The current research contributes to a better understanding of relationships between biotic groups in selected Greek lakes.  相似文献
